Natural Neuroprotective effect from Puerarin a compound in Pueraria lobata in Traditional Chinese Medicine.
Puerarin is the multifunctional plant active ingredient in Pueraria lobata, which has attracted extensive attention due to its potential therapeutic effects on a variety of neurological disorders. Emerging studies have shown that puerarin has different pharmacological effects in animal models of many types of CNS
disorders, including ischemic stroke, SAH, epilepsy, AD, PD, brain trauma, and CNS damage caused by neurotoxic agents.
The clinical use of puerarin as a neuroprotective agent is limited by its low water solubility, poor lipid solubility, and poor intestinal permeability. Various advances in polymer materials, nanotechnology, and particle engineering can solve the problems related to the bioavailability of puerarin. Puerarin loaded nano formulation and puerarin loaded liposomes improve drug stability, bioavailability, and biodistribution to the target. Puerarin has a tendency to interact with biological macromolecules, mainly human serum albumin and low density lipoprotein.
In conclusion, puerarin is a promising natural plant ingredient for treating various CNS disorders. Further structural changes may improve the bioavailability and efficacy of puerarin. Moreover, well-designed, high-quality, large-scale, multicenter randomized clinical studies are needed to determine the safety and clinical utility of puerarin in patients with neurological diseases.
